Research suggests that Andrographis paniculata and its key active compounds — particularly andrographolide and related diterpene lactones — demonstrate meaningful anti-inflammatory activity across a range of laboratory and animal studies, with proposed mechanisms including suppression of NF-κB, MAPK, and JAK-STAT signaling pathways, inhibition of inflammatory markers such as TNF-α, IL-6, IL-1β, COX-2, and iNOS, and more recently documented effects on necroptotic cell death pathways and gut microbiota modulation. Studies indicate that newly identified compounds from the plant, such as andropanilide D, show anti-inflammatory potency exceeding that of andrographolide itself and even the corticosteroid dexamethasone in cell-based assays, while animal models suggest the plant's constituents may preferentially accumulate in inflamed tissue. The available evidence consists almost entirely of in vitro cell studies, computational modeling, and animal experiments, with very few human clinical trials directly evaluating anti-inflammatory outcomes, which substantially limits the conclusions that can be drawn about efficacy in people. Two narrative reviews of traditional Chinese herbs place Andrographis within a broader category of botanicals with plausible anti-inflammatory mechanisms, but both call for more rigorous research before definitive clinical claims can be made.
