The studies reviewed here are largely preclinical — involving mice, fruit flies, cell cultures, and computational models — with no clinical trials represented in this set; one mouse study found that NMN supplementation in aged animals unexpectedly worsened kidney inflammation and injury markers, raising questions about tissue-specific and age-dependent safety, while another computational study using a blood-based gene expression clock reported patterns consistent with a rejuvenating effect on cells in laboratory conditions. Additional studies touched on NMN only tangentially, exploring related NAD⁺ pathway biology in contexts ranging from angiogenesis to viral replication to nerve cell vulnerability, where a related compound (nicotinamide riboside) was shown to have counterproductive effects under certain neurological conditions.
