Research suggests that Lactobacillus rhamnosus GG may support immune function through multiple mechanisms, including modulating cytokine signaling, increasing immunoglobulin levels, promoting T cell development in gut-associated tissues, and activating natural killer cells, with supporting evidence drawn from laboratory cell studies, animal models across several species, and a small number of human trials. Studies indicate that much of the available evidence comes from non-human research — including studies in mice, piglets, foals, and laying hens — which limits how directly the findings can be applied to human health, and the human trials that do exist, including randomized controlled trials examining vaccine immune response and synbiotic combinations in older adults, showed modest or mixed results rather than definitive effects. Some of the more promising human-relevant findings come from a study in older adults where LGG combined with a prebiotic fiber appeared to increase natural killer cell activity and reduce certain inflammatory markers, and from throat spray research suggesting LGG may activate interferon-related immune pathways in upper respiratory tissues. Overall, while the body of research points toward a plausible immunomodulatory role for LGG, most conclusions remain preliminary, strain-specific effects appear to vary meaningfully, and further well-designed human trials are needed before strong conclusions can be drawn.
