L-Proline for Wound Healing

Research suggests that L-proline plays a meaningful role in wound healing, primarily through its function as a key structural precursor to collagen — the protein central to tissue repair and skin integrity. The available evidence comes largely from animal studies and laboratory experiments rather than human clinical trials, and includes rat wound models showing improved tissue regrowth, collagen deposition, and wound strength with both topical and systemic proline administration, as well as in vitro studies demonstrating that incorporating L-proline into biomaterial scaffolds such as chitosan and alginate-gelatin hydrogels significantly accelerates wound closure, supports fibroblast activity, and enhances antioxidant and anti-biofilm properties. A 1965 cell biology study offers foundational mechanistic context, tracing how fibroblasts take up and process proline through intracellular compartments before secreting it as collagen, while a 2016 review highlights hydroxyproline — a modified form of proline found abundantly in collagen — as a promising biomarker for monitoring wound healing and related conditions. The overall direction of the evidence supports a role for L-proline in tissue repair, but the absence of controlled human trials, the variability in delivery methods studied, and the largely preclinical nature of the research base mean that conclusions about its practical use in people remain preliminary.
