The available evidence comes primarily from narrative reviews, laboratory and animal studies, and a smaller number of early-phase human trials, with the overall direction of findings being supportive of I3C's role in modulating estrogen pathways, though direct randomized controlled trial data in humans remains limited. Studies indicate that its metabolite DIM (3,3'-diindolylmethane) shares some of these properties, but one analysis of postmenopausal women on hormone therapy found that DIM supplementation altered estrogen metabolite profiles in ways that could potentially interfere with prescribed estrogen treatments, underscoring that these compounds are biologically active and not without complexity.
