Research suggests that choline and its related compounds — including alpha-GPC, citicoline, and choline bitartrate — play a meaningful role in supporting cognitive function across multiple contexts, with evidence drawn from a 2023 review of preclinical and clinical trials, a meta-analysis of seven randomized controlled trials finding alpha-GPC improved cognition in adults with neurological conditions, and several mechanistic studies pointing to choline's involvement in acetylcholine production, myelin development, and neuroprotection against sleep deprivation and neuroinflammation. Studies also indicate that choline metabolism may be relevant to cognitive resilience in Alzheimer's disease, and that prenatal choline intake could influence long-term brain development in offspring, though a key follow-up study in adolescents is still ongoing and has not yet reported results. However, the evidence is not uniformly positive — a cross-sectional study of over 2,700 older U.S. adults found no significant association between dietary or supplemental choline intake and cognitive test performance, and several of the supportive studies are small, conducted in animals, or focused on specific clinical populations such as those with dementia or neurological disease, limiting how broadly their findings can be applied. Overall, the body of evidence is promising but still developing, with researchers calling for more rigorous longitudinal and experimental studies in healthy human populations before firm conclusions can be drawn.
